我们开发了一种实用的膜蛋白合成策略-可移除增溶骨架修饰策略,能够方便,经济地合成毫克级的中小型膜蛋白。通过在Fmoc固相合成中引入主链碳骨架修饰打破二级结构引起的聚集,亦可引入精氨酸或者赖氨酸助溶标签。该策略使得膜蛋白片段在连接,纯化和质谱表征中行为与普通水溶性多肽几乎相同。我们成功应用该策略制备了甲型流感病毒丝氨酸64位磷酸化的M2质子通道和内向整流钾离子通道蛋白Kir5.1的膜嵌入域,证明了该方法的有效性和实用性。
